The goal of this study is to use eDNA as a cost effective tool for documenting the occurrence and distribution of ESA-listed spring-chinook (Oncorhynchus tshawytscha) throughout the Okanogan and Methow watersheds in an effort to map habitat use and connectivity. Our approach will use eDNA to detect low density spring-chinook in filtered water samples to inventory distribution and re-colonization of tributaries at strategically-sampled points throughout entire stream networks. This approach will provide one of the first applications of eDNA technology for monitoring distribution of native fish species of conservation concern. Stream temperature data will be compiled from federal and provincial government agencies, as well as other data holders in British Columbia which will be housed in an interagency database. Spatial statistical models for river networks like those used for NorWeST will be used with these data to develop a consistent set of high-resolution predictions for all streams and reaches within streams for a pilot area within the Cascadia ecotypic area of the GNLCC (i.e., middle Fraser River and Okanagan River basins). The Confederated Tribes of the Umatilla Indian Reservation (CTUIR) has been working collaboratively with the Bureau of Land Management and US Forest Service (USFS) to inventory and map current and potential distributions of plant communities which support species of cultural concern on federal lands. This proposal is to deliver the pilot component of a wetland tracking project initiated by the CIJV in 2010. The wetland trend project will deliver an assessment of wetland trends and develop an approach for tracking wetland trends into the future; this will have relevance to conservation planning throughout transboundary ecological planning units, including Bird Conservation Regions (BCRs) 9 and 10, and the Cold Deserts and Western Cordillera Ecoregions (Figure 2). Sagebrush (Artemisia spp.)-dominated shrublands are one of the most widespread ecosystems in western North America but also among the most imperiled due to interactions among land use, fire, and exotic plants. Global climate change models predict an accelerated loss of sagebrush due to synergistic feedbacks among disturbance patterns and vegetation response; only 20% would remain under the most extreme scenario of >6 C increase by the end of this century (Fig. 1).
