A large spill of wastewater from oil and gas operations was discovered adjacent to Blacktail Creek near Williston, North Dakota in January 2015. To determine the effects of this spill on streambed microbial communities over time, bed sediment samples were taken from Blacktail Creek upstream, adjacent to, and at several locations downstream from the spill site. Blacktail Creek is a tributary of the Little Muddy River, and additional samples were taken upstream and downstream from the confluence of Blacktail Creek and the Little Muddy River. We evaluated and optimized three rapid methods for extraction of high-quality DNA from carbonate-encrusted microbial communities using modern calcifying oncolites built by cyanobacteria and diatoms in a high-calcium freshwater river. Pulverization, acid (HCl) dissolution, and chelator-mediated (EDTA) dissolution of the carbonate matrix were used and optimized to liberate microbial cells from their mineral encasing. This was followed by cell lysis and DNA extraction and isolation. Fatty acid (FA) analysis is a promising tool to study trophic relationships in soil food webs. We determined FA biomarkers to trace bacterial food sources (Bacillus megaterium, Pseudomonas putida, Enterobacter aerogenes) of Collembola (Heteromurus nitidus, Protaphorura fimata, Folsomia candida). In addition, ?15N, ?13C, C/N ratio, body weight and NLFA/PLFA ratio (neutral lipid/phospholipid fatty acids) of Collembola were assessed. FY2017The Integrated Rangeland Fire Management Strategy Actionable Science Plan places a high priority on assessing control measures for invasive annual grasses, which provide fuel for rangeland fire and impede restoration of desirable perennials. Weed-suppressive strains of the bacterium Psuedomonas fluoresens (P.f.) are an emerging but yet untested tool for selectively reducing these annual grasses.
