Data describe a designed environmental DNA (eDNA) survey for the detection of longnose darter (Percina nasuta) in field collected water samples. Parameters described include the limit of detection and limit of quantification of the assay; a list of freshwater fish species tissue samples that were used to test specificity of the assay; and field collected water samples that were tested to detect the presence of longnose darter DNA. Samples were collected from sites on the St. Francis river, Missouri in 2018.
Catostomid fishes are a diverse family of 76+ freshwater species that are distributed across North America in many different habitats. This group of fish is facing a variety of impacts and conservation issues that are somewhat unique relative to more economically valuable and heavily managed fish species. Here, we present a brief series of case studies to highlight the threats such as migration barriers, flow regulation, environmental contamination, habitat degradation, exploitation and impacts from introduced (non-native) species that are facing catostomids in different regions. The Yellowknife Study Area (YSA), Northwest Territories, Canada, was established in 1961 by H. W. Murdy, U.S. Fish and Wildlife Service, for the study of wetland and waterfowl ecology. The study area is located on the western edge of the Precambrian Shield and edge of the taiga and is recognized for its high wetland densities and waterfowl abundance. The region is underlain by discontinuous, warm permafrost and hence vulnerable to a warming climate. The completion of Highway 3 in early 1960s provided the first access to the region for development, research, and monitoring. The four primary objectives of this project were to: (1) compile a dataset of fish occurrence records for the entirety of the Rio Grande drainage in the US and Mexico; (2) improve that dataset by reformatting dates, synonymizing species names to a modern taxonomy, georeferencing localities, and flagging geographic outliers; (3) for those species with sufficient data for modeling, create species distribution models (SDMs); (4) use the environmental conditions determined via those models to project the species distributions into the future under two climate scenarios. Fish catch and effort data for three species caught in gill nets and trap nets between 1988 and 2019 as part of Minnesota Department of Natural Resources (MNDNR) fisheries surveys conducted during the summer and early fall are included from over 1,300 Minnesota lakes. The three fish species included are: bluegill (Lepomis marochirus) a warm-water adapted species, yellow perch (Perca flavescens) a cool-water adapted species, and cisco (Coregonus artedi) a cold-water adapted species.
