Acanthurid surgeonfishes are an abundant and diverse group of herbivorous fishes on coral reefs. The present study proposes a new methodology for the quantification of parasite eggs in animal tissue. Quantification of parasites are important to understand epidemiology of spirorchiid infections in sea turtles, however different methodologies for quantifying Spirorchiidae eggs in turtle tissues have been used. The most representative way to quantify Spirorchiidae burdens in tissues is counting eggs / g of tissue, however, this method is very laborious. As an alternative, we propose quantifying number of Spirorchiidae egg/ area of tissue on a microscope slide. There is a fundamental knowledge gap on the distribution, prevalence, intensity, and ecology of salmonid myxozoan parasites in the Lake Sammamish watershed, Washington. To address this knowledge gap, we tested water samples for Ceratonova shasta, Parvicapsula minibicornis and Tetracapsuloides bryosalmonae DNA from 84 sites distributed throughout the Lake Sammamish watershed in fall 2019 and 74 sites in spring 2020. Our surveillance identified zones with high waterborne parasite loads and provides a proof of concept for this approach that could be expanded throughout the larger Lake Washington watershed.

Data included are DNA sequences used to identify species boundaries among Ichthyophonus-like parasites. Sequences were generated for three gene regions from each parasite isolate. DNA sequences can be downloaded from GenBank (https://www.ncbi.nlm.nih.gov/) as text or fasta files. Accession numbers for each sequence are included in the data table (.csv file). Data includes 87 sequences (99,383 total nucleotides). Once manuscript is accepted there are no restrictions on dissemination of these data.

We developed and validated conventional and quantitative real-time PCR assays for the detection of DNA from the myxozoan parasite Tetracapsuloides bryosalmonae, the causative agent of proliferative kidney disease in fish. Assays were tested on fish tissue and on field-collected water samples to assess diagnostic and environmental DNA capabilities. The specificity, sensitivity, and broad applicability of the present assays surpass previous methods for detecting T. bryosalmonae DNA from fish tissue and water samples.

Despite being one of the most prevalent forms of consumerism in ecological communities, parasitism has largely been excluded from food-web models. Stable isotope analysis of consumers and their diets has been widely used in the study of food-webs for decades. Data are relative fluorescent units obtained from the FAM channel of a AmpliFire Isothermal Fluorometer (Agdia, Inc., Elkhart, USA) recorded every 10 sec during as isothermal (LAMP) amplification. The isothermal reaction is to detect the Asian fish tapeworm, Schyzocotyle acheilognathi [syn. Bothriocephalus]. A probe-based quantitative real-time PCR assay was developed to detect Tetracapsuloides bryosalmonae, which causes Proliferative Kidney Disease in salmonid fish, in kidney tissue and environmental DNA (eDNA) water samples. Environmental DNA detection results from samples collected using autonomous water sampling robots and manual approaches. Samples were collected in the Upper Yellowstone River (Montana) and Upper Snake River (Idaho/Wyoming) in 2018 and 2019. Samples were tested for the DNA of the following species: the waterborne protozoa Naegleria spp., the fish pathogen Tetracapsuloides bryosalmonae, Scomber japonicas (mackerel fish), kokanee salmon (Oncorhynchus nerka) and dreissenid mussel (Dreissena spp.).

We characterized a natural sea louse epizootic of Caligus clemensi and the effects of parasitism on Pacific herring Clupea pallasii in Port Angeles Harbor, WA, USA. Infestation prevalence on newly metamorphosed age0 Pacific herring reached 100% prevalence by mid-August. At this time, the mean louse intensity was 4.6 lice and a positive correlation occurred between louse intensity and Pacific herring body length. The epizootic then waned, with infestation prevalence decreasing to less than 25% and the mean parasite intensity falling below 1 louse by October.
