Biological indicators including morphometric (length, weight), age, sex and health indicators including organismal (health assessment index, condition factor), organ (hepatosomatic index, gonadosomatic index), cellular (intersex, parasite density, macrophage aggregate density), subcellular (plasma vitellogenin, estradiol, testosterone, 11-keto testosterone) and molecular (hepatic transcript abundance) were measured in smallmouth bass from tributaries in the Chesapeake Bay watershed. Total mercury was measured in these fish and correlations between mercury and biological indicators were assessed.

Decades of poor reproductive success and young-of-the-year recruitment, in addition to adult mortality, has led to a decline in the smallmouth bass (SMB) population in subwatersheds of the Potomac River. Previous studies have identified numerous biologic and environmental stressors associated with negative effects on SMB health. To better understand the impact of these stressors, the current study was conducted from 2013-2019 to identify temporal changes associated with SMB reproductive health. Grab surface water samples were collected and analyzed for over 300 organic contaminants, including pesticides, phytoestrogens, pharmaceuticals, hormones and total estrogenicity (E2Eq). Adult SMB were collected and sampled...

Histopathological assessments of young-of-the-year smallmouth bass (YOY SMB) in the Susquehanna River drainage identified a high prevalence of the myxozoan Myxobolus inornatus. This myxozoan infects the connective tissue of the muscle below the skin but is sometimes observed in the esophagus and buccal cavity. In some instances, shallow infections cause breaks in the skin which could increase the chance of opportunistic bacterial infections. Several microbial pathogens including Flavobacterium columnare, Aeromonas spp. and largemouth bass virus (LMBV) have also been cultured from clinically diseased YOY. A multiplex fluorescence in situ hybridization (FISH) assay was developed to determine potential co-localization...

Largemouth base histological development and transcriptomic changes in gonad tissue after early life stage exposure to Atrazine (1-Chloro-3-ethylamino-5-isopropylamino-2,4,6-triazine) or the model estrogen 17α-ethinylestradiol.
Monitoring wild fish health and associated exposure effects in regions where agriculture and development have impacted rivers and streams has become a valuable research tool. Smallmouth bass (Micropterus dolomieu) are a sensitive, indicator species that exhibit symptoms of immunosupression and endocrine disruption in response to water quality changes and contaminant exposure. In the Susquehanna River drainage, smallmouth bass have been used in biomonitoring assessments to help understand changes in the population due to poor young-of-the-year recruitment and health effects. To help determine effects of agriculture and development on smallmouth bass health, two sites (a developed/agriculture site and a forested site)...

In 2016, smallmouth bass were collected from three sites in the Lake Erie drainage in Erie County, Pennsylvania and an out-of-basin site in the Allegheny River drainage in order to assess reproductive health and identify endocrine disruption responses. Pieces of liver and gonad were taken in Z-Fix and RNALater for histology and transcript abundance analysis. A landscape analysis was conducted for the immediate and upstream catchments with 2016 land cover data. Correlations between biological variables and transcripts were made in addition to differences between sites. The results showed that males had biological variables that correlated with transcripts associated with endocrine disruption and a high rate of testicular...

This data release includes laboratory analyses for fish tissue samples collected from Lake Koocanusa and the Kootenai River Basin in Montana during 2022. Samples were collected by the U.S. Geological Survey Wyoming-Montana Water Science Center in cooperation with Montana Fish, Wildlife and Parks and Montana Department of Environmental Quality. Analyses include histology, percent moisture, and 30 elements, including selenium, mercury, and methylmercury. This data release also compiles laboratory quality-assurance and quality-control data associated with the samples.
This dataset includes fish scientific name, fish common name, site identification codes, latitude, longitude, site name, sample identification codes, collection date, tissue type, percent moisture content in fish samples, and wet and dry total mercury concentrations in fish tissues.

There is an increasing emphasis on effects-based monitoring to document responses associated with exposure to complex mixtures of chemicals, climate change, pathogens, parasites and other environmental stressors in fish populations. For decades aquatic monitoring programs have included the collection of tissues preserved for microscopic pathology. Consequently, formalin-fixed, paraffin-embedded (FFPE) tissue can be an important reservoir of nucleic acids as technologies emerge that utilize molecular endpoints. Despite the cross-linking effects of formalin, its impact on nucleic acid quality and concentration, amplification, and sequencing are not well described. While fresh-frozen tissue is optimal for working with...

Intersex in small- and largemouth bass has been observed in the upper Chesapeake Bay Watershed since 2003. This observation is indicative of exposure to estrogenic endocrine disrupting chemicals. In order to complement biological data sets and better identify the spatial and temporal nature of this end-point, discrete grab water samples were collected throughout the upper Chesapeake Bay Watershed for the evaluation of estrogenic activity. Data reported here are estrogenicity values derived from bioassay screening of OASIS-HLB extracts discrete grab water samples. Estrogenicity was determined using a bioluminescent yeast estrogen screen. Here we report estrogenicity data for selected sites in the Chesapeake Bay water...

Archived plasma from fish health monitoring sites within the Chesapeake Bay watershed was analyzed for 13 perfluoroalkyl compounds. Smallmouth bass were collected from two sites in the Potomac River drainage (Antietam Creek, Maryland and South Branch Potomac, West Virginia) and two in the Susquehanna River drainage (West Branch Mahantango Creek, Pennsylvania and Pine Creek, Pennsylvania). Site and date of collection, length, weight and age of each adult smallmouth bass were documented. Four compounds were detected in every plasma sample, while two additonal compounds were only occasionally observed at low concentrations.

Adult and young of the year smallmouth bass from the Chesapeake Bay watershed have faced health issues (mortality events, disease by a variety of pathogens, endocrine disruption in male fish, evidence of oxidative damage to tissues, and signs of exposure to chemical contaminants) at multiple sites within the Susquehanna and Potomac River basins. Functional immune assays were added to long-term comprehensive monitoring assessments to determine if immunosuppression was playing a role in observed disease and mortalities because no one single or consistent cause could be identified. The functional immune assays measured bactericidal activity, respiratory burst activity, and lymphocyte mitogenesis using leukocytes isolated...

The data were gathered as a preliminary assessment of estrogenicity under base-flow conditions at over 100 sites (lakes and streams) throughout New Jersey followed by more targeted sampling of smallmouth bass at nine sites with varying levels of estrogenicity. In 2016, 102 sites (lakes and streams) for the preliminary estrogenicity assessment were selected utilizing compiled results of previous monitoring studies (chemical and biological), current sampling networks, and other areas of concern based on input from stakeholders. Discrete grab surface water samples were collected under base-flow conditions in the fall of 2016 and analyzed for levels of estrogenicity using a bioluminescent yeast screen. Water samples...
