The present study proposes a new methodology for the quantification of parasite eggs in animal tissue. Quantification of parasites are important to understand epidemiology of spirorchiid infections in sea turtles, however different methodologies for quantifying Spirorchiidae eggs in turtle tissues have been used. The most representative way to quantify Spirorchiidae burdens in tissues is counting eggs / g of tissue, however, this method is very laborious. We developed and validated conventional and quantitative real-time PCR assays for the detection of DNA from the myxozoan parasite Tetracapsuloides bryosalmonae, the causative agent of proliferative kidney disease in fish. Assays were tested on fish tissue and on field-collected water samples to assess diagnostic and environmental DNA capabilities. The specificity, sensitivity, and broad applicability of the present assays surpass previous methods for detecting T. bryosalmonae DNA from fish tissue and water samples.

There is a fundamental knowledge gap on the distribution, prevalence, intensity, and ecology of salmonid myxozoan parasites in the Lake Sammamish watershed, Washington. To address this knowledge gap, we tested water samples for Ceratonova shasta, Parvicapsula minibicornis and Tetracapsuloides bryosalmonae DNA from 84 sites distributed throughout the Lake Sammamish watershed in fall 2019 and 74 sites in spring 2020. Our surveillance identified zones with high waterborne parasite loads and provides a proof of concept for this approach that could be expanded throughout the larger Lake Washington watershed.

Data included are DNA sequences used to identify species boundaries among Ichthyophonus-like parasites. Sequences were generated for three gene regions from each parasite isolate. DNA sequences can be downloaded from GenBank (https://www.ncbi.nlm.nih.gov/) as text or fasta files. Accession numbers for each sequence are included in the data table (.csv file). Data includes 87 sequences (99,383 total nucleotides). Once manuscript is accepted there are no restrictions on dissemination of these data.

Monitoring wild fish health and associated exposure effects in regions where agriculture and development have impacted rivers and streams has become a valuable research tool. Smallmouth bass (Micropterus dolomieu) are a sensitive, indicator species that exhibit symptoms of immunosupression and endocrine disruption in response to water quality changes and contaminant exposure. Samples were collected in the Upper Yellowstone River (Montana) and Upper Snake River (Idaho/Wyoming) in 2018 and 2019.
